ELISA assays are assays where there is a need to measure the presence and amount of an antibody to a particular molecule. These circulating antibodies can be present because the animal or person has been given a particular “foreign” antigen or are self-generated auto-antibodies. ELISAs involve the capture of specific circulating antibodies by antigen immobilized onto the solid phase. The captured antibody is then quantified or determined by addition of either a general isotype antibody conjugate, i.e., HRP-labeled antibody to human IgG; or a more specific subtype conjugate, such as a HRP-labeled antibody to IgG1. Arbor Assays will not develop ELISAs to infectious antigens.
We can carry out the stages of this work including the conjugation of the peptide, nucleic acid or protein for antibody isolation. This allows the generation of a standard for the assay. After the antigen has been immobilized we will select a vendor to process the sera for antibody isolation. The vendor will carry out the basic isolation and characterization of the antibody.
Based upon your specifications we can immobilize the antigen to the solid phase, formulate the isolated standard antibody, carry out the assay optimization to generate an appropriate standard curve and complete the validation process. We can develop an isotype assay to identify IgG, M, A, etc and/or develop a specific subtype assay or range of assays. The readout for your assay can be selected from traditional HRP or AP enzymatic colorimetric detection to fluorescent, chemiluminescent or bioluminescent. We also have experience of developing microparticle based assays from the ground up. Our validation methods are stringent and complete.
We will undertake as many projects or as much of a project as you wish us to undertake. We have the capacity to develop, validate and manufacture the entire assay if you so request. We are not a GMP/GLP facility at this time but the founders have worked in all of these aspects at both life sciences companies and within the FDA regulated diagnostics industry.
Notes on Services
Please remember that we are only as good as the information we receive from you, our customer. We need a well-defined idea of what you would like made. Typically that only happens after we have a signed non-disclosure agreement (NDA). The NDA can be either a one-way, or more preferable, a two-way version. After the NDA has been put in place you should tell us everything you know about the molecule and everything you want to get from the product we’re building. This information allows us to complete the contractual work rapidly and at least cost to you.
We typically carry out projects on a time and materials basis. After adequate discussion we can generate an assessment of the amount of time that will be required to complete the project. You only pay for actual Arbor Assays scientist time, plus any special raw materials such as custom antibodies, enzymes, or outside synthetic work. Again, please remember, you do not pay for our services when the rabbits, mice, goats or chickens are generating antibodies, or when one of our contract synthetic companies are making peptides or organic conjugates. You only pay for our work time.
