Ann Arbor, MI (November 14, 2019) — Arbor Assays adds two new potent STING inhibitors, H-151 and DMXAA, to their catalog of inflammation kits and reagents. The addition of the STING inhibitors combined with Arbor Assays 2’,3’-Cyclic GAMP ELISA Kit provides important new tools for biomedical research and drug discovery.

H-151 is an irreversible inhibitor of Stimulator of Interferon Genes (STING) in mouse and human cells. H-151 covalently binds to Cys91 of STING, preventing activation via blockade of palmitoylation and clustering of STING. H-151 blocks STING-induced expression of pro-inflammatory cytokines and reduces inflammation in models of auto-inflammatory disease.

DMXAA (Vadimezan) is a STING agonist selective for mouse STING. Intratumoral administration of DMXAA results in tumor regression and complete rejection in mouse xenografts. DMXAA administration a cascade of cellular events that includes disruption of tumor vasculature and release of chemokines triggering the recruitment of immune cells. DMXAA induced expression of IFN-β results in a striking expansion of leukemia-specific T cells extending survival in two acute myeloid leukemia models.

The Arbor Assays 2’,3’-Cyclic GAMP ELISA Kits accommodate a wide variety of cell lysis buffer types and biological matrices. The kit uses the customer’s preferred cell lysis buffer either directly or diluted into Assay Buffer containing EDTA to minimize the activity of Zn2+-activated hydrolyses capable of cleaving 2’,3’-Cyclic GAMP molecules in some samples. The cGAMP ELISA Kits can measure cGAMP in cell lysates, tissue extracts, and tissue culture media. The kits are highly specific for cGAMP, have a wide dynamic range (20 – 0.082 pmol/mL), and excellent sensitivity (LLOD = 0.048 pmol/mL).
