Assay Principle: 

The DetectX® Prostaglandin E2 (PGE2) ELISA Kit is designed to quantitatively measure PGE2 present in saliva, urine, serum, plasma, and tissue culture media samples. This assay is a competitive enzyme-linked immunosorbent assay (ELISA).

Standards or diluted samples are pipetted into a clear microtiter plate coated with goat anti-mouse IgG. A PGE2–peroxidase conjugate and a monoclonal antibody specific for PGE2 are added to each well. Free PGE2 in the sample competes with the PGE2–peroxidase conjugate for antibody binding. After incubation, the plate is washed and tetramethylbenzidine (TMB) substrate is added. The substrate reacts with the bound PGE2–peroxidase conjugate to generate a colorimetric signal. The intensity of the signal is inversely proportional to the concentration of PGE2 in the sample and is measured at 450 nm using a microplate reader.

PGE2 concentrations in unknown samples are determined by comparison to a standard curve generated using known concentrations of PGE2, with appropriate correction for sample dilution.

Background:

Prostaglandin E2 (PGE2), a critical member of the prostaglandin family, plays a vital role in various physiological and pathological processes. Prostaglandins are synthesized from arachidonic acid via the cyclooxygenase (COX) enzymes. COX-1 and COX-2 convert arachidonic acid into PGH2, which is then processed into PGE2 and other prostanoids.

PGE2 is known for its involvement in inflammatory responses and is present in various tissues and pathological conditions such as arthritis, fever, tissue injury, endometriosis, and multiple cancers. Its biological actions include vasodilation, modulation of sleep/wake cycles, stimulation of bone resorption, and thermoregulatory effects. PGE2 is also significant in renal physiology, impacting sodium excretion and renal hemodynamics.
