Wildfire Smoke and Male Fertility: Insights from Recent Research 

Wildfires are becoming more frequent and severe across many regions of the world. While the respiratory and cardiovascular effects of smoke exposure are well documented, researchers are only beginning to understand how wildfire smoke may affect reproductive health. 

Recent studies have linked wildfire smoke exposure to impaired male fertility in firefighters. To better understand these observations, scientists used the Arbor Assays DetectX® Corticosterone Multi-Format ELISA Kit (K014-H1) alongside assessments of reproductive hormones and sperm quality in a rat model of episodic eucalyptus smoke exposure. 

Published in Environmental Science & Technology, the study found that just seven days of smoke exposure during sperm maturation reduced multiple measures of sperm motility, providing new evidence that wildfire smoke may impact male reproductive health. 

Investigating a Sensitive Window in Sperm Development 

The research team focused on sperm maturation, a critical stage of development that occurs in the epididymis. During this process, sperm acquire the motility required for successful fertilization. 

Researchers focused on sperm maturation because it occurs over approximately two weeks in both rats and humans, making it a useful model for studying the effects of short-term wildfire smoke exposure. 

Twelve-week-old Long-Evans rats were exposed to eucalyptus smoke for seven one-hour sessions over ten days. Smoke concentrations ranged from 3.52 to 23.7 mg/m³, and an additional group was exposed to filtered smoke that removed most particulate matter while retaining volatile compounds. Researchers then collected blood samples and assessed sperm motility approximately 24 hours after the final exposure. 

As part of the hormonal assessment, plasma corticosterone concentrations were measured using the Arbor Assays DetectX® Corticosterone ELISA Kit. Corticosterone is the primary glucocorticoid stress hormone in rodents and is commonly used to assess physiological stress responses. 

Smoke Exposure Reduced Sperm Motility 

The researchers observed measurable declines in sperm performance following smoke exposure. At an exposure concentration of 11.0 mg/m³, rats exhibited a 6% reduction in progressive sperm tracks and a 5% reduction in average path velocity (VAP), both of which are important indicators of sperm motility. 

Perhaps more surprisingly, the filtered smoke group also showed significant effects. Rats exposed to smoke with particulate matter removed experienced: 

  • 14–15% reductions in progressive motility and tracking 
  • 9–12% reductions in sperm velocity measurements  
  • An 8% reduction in the amplitude of lateral head displacement  

These findings suggest that volatile compounds present in smoke may contribute substantially to reproductive toxicity. In fact, the filtered smoke exposure often produced larger effects than whole smoke exposure, leading the researchers to conclude that the gaseous phase of smoke warrants further investigation as a driver of fertility-related outcomes. 

Corticosterone Helped Provide Biological Context 

One of the most interesting findings was what corticosterone data did not show. 

Despite the reductions in sperm motility, corticosterone levels remained largely unchanged in the lower-concentration and filtered-smoke groups, where many of the strongest reproductive effects were observed. The consistent corticosterone levels suggest that impaired sperm function was not simply the result of a generalized stress response.
